Based in Wetherby, my client is a preclinical phase market leader in the next generation of Biotherapeutics to meet the growing demand for the next generation of drugs that will compete with monoclonal antibodies and other antibody mimetics, primarily in oncology. The expansion of their research activities at Wetherby has created an opportunity for a scientist within their production team.
You will be a member of the Production team responsible for the expression and purification of proteins for internal and customer facing projects. This mainly involves E. coli expression but the team will also need to use other expression modalities. The team handles all aspects of protein production from cell culturing through induction, growth monitoring, and cell harvesting to material processing, chromatography (preparative and analytical) and SDS-PAGE. The team are also expected to be able to use molecular biology techniques to re-clone constructs to produce alternative fusion proteins as required for specific projects.
+ Express and purify proteins for internal and custom projects using a range of different expression platforms (e.g., bacterial, yeast or mammalian cells).
+ Characterisation of proteins using a range of methods (HPLC, SDS-PAGE, western blot, ELISA, Optim, SPR, etc).
+ Carry out optimisation studies to increase production from host cells.
+ Produce high quality proteins and tool molecules for internal or custom projects, delivering within timelines and carrying out the QC to deliver the correct specification.
+ Develop purification protocols and processes using conventional chromatography resins to purify up-to-gram quantities of proteins on ÄKTA systems.
+ A PhD + 1 or more years working in the field of biotechnology or MSc + 3 or more years working in the field of biotechnology or 5 or more years working in the field of biotechnology.
+ Standard protein production techniques and protein engineering knowledge, including working with a range of protein expression platforms (e.g., bacterial and mammalian).
+ Strong practical knowledge of the ÄKTA purification platform, Unicorn software and using conventional chromatography resins e.g., HIC, ion exchange, multimodal etc., to build multistage protein purification processes to achieve high recovery and purity yields of product.
+ Excellent knowledge of a range of methodologies to characterise proteins (e.g., SPR, ELISA, SDS-PAGE, Western blot).
